Title: A new role for interferon gamma in facilitating senescence escape in melanoma
Abstract: Melanoma is the deadliest form of skin cancer. For the second most common subtype, NRAS-mutated, recent clinical trials assess a new therapeutic approach, the combined inhibition of the cell cycle and MAPK pathway. We treated 3 cell lines for 33 days and characterized cellular responses by acquiring different omics data at multiple time points. We investigated changes in transcriptome, kinome activity and miRNA interactome. Finally, we integrated bulk RNA-seq and in house single cell data though an NMTF-based method called “iCell”. Overall, our data reveal that besides distinct cellular phenotypes, all cell lines trigger a senescent program and interferon responses. Our findings suggest a new role of interferon gamma in senescence escape and predict novel genes involved.
